Rein Roos has studied percussion at Tallinn School of Music under the guidance of Rein Saue (1970–1972). In 1977 Rein Roos graduated from Tallinn State Conservatoire under the guidance of Matti Reimann and Kalju Terasmaa.

Since 1971 Rein Roos is percussionist at Estonian National Symphony Orchestra. He has worked also at Estonian National Opera Symphony Orchestra (1981–1991 and 1995–1997). In 1984–1999 Rein Roos was percussion pedagogue at Tallinn Music High School. Since 1994 Rein Roos is pedagogue at Georg Ots Tallinn Music School and since 1999 also at Estonian Academy of Music and Theatre (since 2004 docent). Rein Roos’ students include among others Anto Õnnis, Heigo Rosin, Anneli Kadakas, Andrus Lillepea, Vambola Krigul, Hele-Riin Uib, Aleksandra Anstal, Lehari Kaustel, Martin Sildos, Kristjan Mäeots, Madis Metsmart and Kaspar Eisel.

Rein Roos has participated also at orchestras as NYYD Ensemble and Tallinn Chamber Orchestra. He has participated at recordings and has been first performer of several Estonian composers works including Alo Põldmäe, Raimo Kangro, Urmas Sisask and Toivo Tulev.
